Surface Layer of Wool. I. Dityrosine Synthesis and Characterization.

Research on the surface layer of wool performed an assay of the enzymatic oxidation of tyrosine to dityrosine to establish the optimal conditions for dityrosine synthesis. Modifications to the procedures for dityrosine synthesis and purification resulted in a higher yield (10 percent) of pure dityrosine. Modifications included the use of type II peroxidase enzyme, an increase in the tyrosine solubility, preheating the tyrosine solution, and the use of activated carbon and gel filtration chromatography. The pure dityrosine served as a reliable reference for research on the dityrosine content of wool. The presence of dityrosine in wool keratin probably promotes mechanical stability of the protein by providing a covalent crosslinking between peptide chains. 15 refs.
