high pressure-temperature bioreactor: assays of thermostable hydrogenase with fiber optics

elucidating structure-function relationships for certain enzymes requires specialized equipment for studying the kinetics of enzyme-catalyzed reactions under extreme conditions. a novel bioreactor is described which is suitable for measuring enzyme activity uv-visible spectrophotometry or gas chromatography at temperatures up to 260 $degree$ c and pressures up to 667 atm experiments indicate that the growth rate of methanococcus jannaschii at both 86 and 90 $degree$ c was enhanced by hyperbaric pressures of he up to 750 atm. at each temperature the rate of methanogenesis was doubled by an increase in pressure from 7.8 to 250 atm. although increasing the pressure to 250 atm did not extend the upper temperature limit for growth, it did increase the upper temperature limit for methanogenesis by at least 5 $degree$ c. 22 refs
