penicillinase is immobilized on a single fiber-optic ph sensor by cross-linking in a thin glutaraldehyde membrane on a porous glass bead, to which a fluorescent ph dye has been bound. pencillin in solution is enzymatically converted to penicilloic acid at the probe surface, causing a decrease in ph with a concomitant decrease in the fluorescent intensity of the dye. an argon ion laser is used to excite fluorescence and the fluorescent radiation is collected in the same fiber for transmission to the detector via a coupling device the signal reaches 95 of total response in 20-45 s. the detection limit is 0. 1 mm penicillin.
