Ezymatic Hydrolysis Cellulose: Analysis by Use of Diffusion Equations

Because cellulose is an insoluble substrate, many kinetic equations used for enzymatic reactions in solution might be invalid. A novel approach to characterize the hydrolysis of cellulose by a cellulase mixture measured product formation at different flow rates for extended periods. A series of experiments measured weight loss and percent hydrolysis during hydrolysis of cellulose by cellulase and calculated rate constants and diffusion coefficients according to the diffusion equation solutions of Hill and Newman. Hill's equation was applicable to systems without flow dependent boundary layers, whereas Newman's equation was applicable when significant boundary layer effects were present. The application of diffusion equations revealed useful mechanistic information not obtained by Michaelis - Menten Kinetics. Abstract.
