THANNIN INHIBITS INTRACELLULAR CALCIUM AND cAMP PATHWAYS.

Tannin, isolated from cotton bracts, inhibits chloride and water secretion across the airway epithelium. Since intracellular calcium (Ca) and cAMP levels regulate chloride secretion in airway epithelial cells, the effect of cotton bracts tannin on intracellular Ca in response to epinephrine and bradykinin stimulation, and its relationship to intracellular levels of cAMP in bovine trachea epithelia cells, were examined. Indo-1 fluorescent techniques were used to determine levels of intracellular Ca, and a radiolmmunassay kit was used to determine intracellular cAMP levels. Tannin had no effect on intracellular Ca or cAMP levels in response to bradykinin. In contrast, tannin inhibited the epinephrine-induced rise in both intracellular Ca and cAMP levels. Tannin may inhibit chloride secreation by affecting intracellular Ca and cAMP pathways, but not prostagiandin pathways. 27 refs.
