the induced circular polarized fluorescence (icpf) spectra and the induced anisotropy factors for absorption and fluorescence are measured to find out how the dye binding in an electronically excited state of an alpha-chymotrypsin (alphact)-dye complex differs from that in its ground state. two types of icpf spectra are found: one is a single negative cpf band which is due to a 1'1 alphact-dye complex (dye' proflavine (pf), acriflavine (af)) and the other consists of one negative and one positive cpf bands which are mainly due to a complex of the composition, one alphact to two dye molecules (dye' rhodamine 6 g (rg), pyronin (pb)). it is shown that the observed change of sign in the icpf and the induced circular dichroism spectra and also in the fluorescence and absorption anisotropy factors in the former case can be explained if the relative position of pf (af) and chromophores in the active-site environment of alphact in an excited-state complex slightly differs from that in the ground-state complex
